Her show that was a one-woman production, The Patriots (a sketch show created by her) premiered in November 2005 at the Upright Citizens Brigade Theatre. She was awarded the Breakout Performer Award at HBO U.S. Comedy Arts Festival in Aspen, Colorado in March 2006. Coupe was married from December 2007 until June 2013 to Randall Whittinghill (acting teacher, puppeteer). She announced her engagement to Darin Oliveren on Instagram on November 27 in 2014. While on vacation in New Zealand, they were married on December 24, 2014. In 2018, they filed for divorce. Between April 2011 and May 2013, Coupe was a guest on ABC's ensemble comedy series Happy Endings, along with Zachary Knighton (Adam Pally), Damon Wayans Jr., Casey Wilson, and Zachary Knighton. The show was cancelled by ABC on May 3, 2013, even though it received acclaim from critics. It also gained an audience of cult. Following the time that Happy Endings was cancelled, she appeared as a guest in the third season of House of Lies. She also appeared alongside Jay Harrington on the comedy series Benched. It premiered on October 28th, 2014. Then she was cast as the character of Hannah Wyland in Quantico, an ABC thriller.
